To achieve playback of m.20 grace notes do this: Options > Play > As Recorded. Also you have slurs that don’t make sense in that measure.

Double click on your Grand Pause then make your preferred settings. Make sure that you’ve ticked the Play box at the top left of the Edit Playback window.

Not sure if this is what you want, but open your Tools Panel, go to Bar Lines, go to Cross Staff Barlines then draw a narrow rectangle with your mouse that encompasses one entire system and your barlines will be properly completed throughout your entire score. Latency, audio settings, memory, disk access speeds (HD vs SSD) are all worth investigating as possible culprits. Somewhere between a buffer size of 2816 and 0 you should be able to find, if not a perfect spot, at least a ‘sweet spot’.

Once you’ve confirmed that ASO is up and running through the Kontakt engine downloading and running this file will load and play the ASO sounds. Your version of Overture 5.0.4 goes way back to April 16, 2016. Perhaps there is an incompatibility between the newest Windows 10 and this older version of Overture.
